Integrated Bioinformatics Approach for Disclosing Autophagy Pathway as a Therapeutic Target in Advanced KRAS Mutated/Positive Lung Adenocarcinoma

چکیده

Background: Lung cancer is the leading cause of cancer-related deaths, accounting for 1.8 million deaths (18%). Nearly 80%-85% lung cases are non-small cell cancers (NSCLC). One most frequent genetic mutations in NSCLC Kirsten Rat Sarcoma Oncogene Homolog (KRAS) gene mutation. In recent years, autophagy has drawn substantial attention as a potential pathway that can be targeted driven by KRAS mutation to efficiently improve therapeutic profile different treatments. Methods: this study, we have investigated targeting treatment approach advanced KRAS-mutated adenocarcinoma using expression data from The Cancer Genome Atlas Adenocarcinoma (TCGA-LUAD) project. Results: Compared wild-type adenocarcinoma, there were found 11 differentially expressed autophagy-related genes (DEARGs), with 5 upregulated and 6 downregulated DEARGs (threshold adjusted p-value <0.05). Conclusion: These inhibitors.

KRAS as a Therapeutic Target.

KRAS proteins play a major role in human cancer, but have not yielded to therapeutic attack. New technologies in drug discovery and insights into signaling pathways that KRAS controls have promoted renewed efforts to develop therapies through direct targeting of KRAS itself, new ways of blocking KRAS processing, or by identifying targets that KRAS cancers depend on for survival. Although drugs ...

Bioinformatics-Based Prediction of FUT8 as a Therapeutic Target in Estrogen Receptor-Positive Breast Cancer

Abstract Introduction: Estrogen receptor-positive (ER-positive) breast cancer is a subgroup of breast tumors that is more likely to respond to hormone therapy. ER-positive and ER- negative breast cancers tend to show different patterns of metastasis because of different signaling cascade and genes that are activated by estrogen response. Genetic factors can contribute to high rates of metastas...

Overactivated neddylation pathway as a therapeutic target in lung cancer.

BACKGROUND A number of oncoproteins and tumor suppressors are known to be neddylated, but whether the neddylation pathway is entirely activated in human cancer remains unexplored. METHODS NEDD8-activating enzyme (NAE) (E1) and NEDD8-conjugating enzyme (E2) expression and global-protein neddylation were examined by immunohistochemistry, immunoblotting, and real-time polymerase chain reaction a...

Autophagy: a potential therapeutic target in lung diseases.

Macroautophagy (hereafter referred to as autophagy) is an evolutionally conserved intracellular process to maintain cellular homeostasis by facilitating the turnover of protein aggregates, cellular debris, and damaged organelles. During autophagy, cytosolic constituents are engulfed into double-membrane-bound vesicles called "autophagosomes," which are subsequently delivered to the lysosome for...
